Kinetics of reaction of 2-bromoacetamide with a 39 base pair duplex DNA sequence was studied. | [Synthesis]
Step 1: To a dichloromethane solution containing the amine (300 mg, 0.71 mmol) and potassium carbonate (290 mg, 2.1 mmol) obtained from Step 1 of Example 51 was slowly added bromoacetyl chloride (71 μL, 0.85 mmol) at room temperature. The reaction mixture was stirred at room temperature overnight. Upon completion of the reaction, the reaction mixture was washed with 0.1 N NaOH aqueous solution and subsequently dried over anhydrous sodium sulfate. The solvent was removed by concentration under reduced pressure and the residue was purified by silica gel column chromatography (eluent: dichloromethane/ethyl acetate, 9:1, v/v) to afford the target product 2-bromoacetamide 281 mg in 73% yield. | [References]
