GENERAL STEPS: At room temperature, o-methylacetophenone (0.5 mmol), thiourea (0.5 mmol) and triethylamine (0.5 mmol) were dissolved in acetonitrile (3 mL) in a round bottom flask. Subsequently, carbon tetrabromide (0.5 mmol) was added to the reaction system and the reaction was initiated with stirring. The reaction lasted from 2 to 6 hours, during which the progress of the reaction was monitored by thin layer chromatography (TLC). Upon completion of the reaction, water (5 mL) was added to the mixture and extracted with ethyl acetate (3 x 5 mL). The organic phases were combined, dried with anhydrous magnesium sulfate, filtered and concentrated under reduced pressure to obtain the crude product. Finally, the crude product was purified by silica gel column chromatography using a gradient mixture of hexane/ethyl acetate as eluent to obtain analytically pure 2-amino-4-o-tolylthiazole. | [References]
