2-Amino-6-chloropyridine (15 g, 116 mmol) was dissolved in chloroform (600 ml) at 0°C and a solution of bromine (4.2 g, 965 mmol) in chloroform (50 ml) was added slowly and dropwise. After the dropwise addition, the reaction mixture was warmed up to room temperature and stirred continuously for 16 hours. Upon completion of the reaction, the reaction was quenched with ice-cold water followed by extraction of the organic layer with dichloromethane (DCM). The organic layers were combined and concentrated to give the crude product. The crude product was purified by silica gel column chromatography using hexane solution of 10% ethyl acetate as eluent to give 2-amino-3-bromo-6-chloropyridine (6.2 g, 25.5% yield).LCMS analysis showed the molecular ion peak of the target compound m/z = 209.0 ([M + H]+). | [References]
