Example 2.5 Synthesis of 2,3-diamino-5-fluoropyridine: 2-amino-5-fluoro-3-nitropyridine (0.42 g, 2.3 mmol) and iron powder (1.6 g, 28 mmol) were dissolved in ethanol (10 mL), followed by addition of water (0.5 mL, 28 mmol) and hydrochloric acid (27 μL, 0.32 mmol). The reaction mixture was heated and refluxed for 1 hour. After that, iron powder (0.2 g, 3.6 mmol) was added additionally and reflux was continued for 30 min. Upon completion of the reaction, the mixture was filtered through diatomaceous earth and the filtrate was concentrated under reduced pressure to give 0.3 g of crude product (100% yield). The product was characterized by 1H NMR (300 MHz, CDCl3): δ 3.55 (broad peak, 2H), 4.1 (broad peak, 2H), 6.7 (double peak, 1H), 7.5 (single peak, 1H). | [References]
