GENERAL STEPS: Benzyl bromide (0.052 mL, 0.6 mmol) was added to a mixture of 6-chloropurine (0.056 g, 0.5 mmol) and sodium hydroxide (0.014 g, 0.6 mmol) in deionized water (2 mL). The reaction mixture was placed in a microwave reactor and irradiated at 0-400 W power (internal temperature maintained at 100 °C) for 10 min. Upon completion of the reaction, the mixture was concentrated under reduced pressure to remove the solvent and the residue was purified by column chromatography using ethyl acetate-cyclohexane (9:1, v/v) as eluent to give 9-benzyl-6-chloro-9H-purine in 82% yield. | [References]
