In a 10 mL glass tube, 2-iodo-5-methoxybenzoic acid (1.0 mmol), formamidine hydrochloride (1.2 mmol), CuCl2 (0.1 mmol), ligand L2 (0.1 mmol), and NaOH (4.0 eq.) were added sequentially, followed by 3.0 mL of water. After sealing the reaction vessel with a septum, it was placed in a microwave reactor. Using a CEM Discover microwave synthesizer, microwave radiation was applied at 120 W for 20 min at room temperature, during which the reaction mixture was kept continuously stirred. After completion of the reaction, the solvent was removed by distillation under reduced pressure. The crude product was purified by silica gel column chromatography to afford 6-methoxy-4(1H)-quinazolinone. All products were analyzed by nuclear magnetic resonance (NMR) and mass spectrometry (MS) to confirm their structures. | [References]
