General procedure for the synthesis of 7-chlorofuro[3,2-b]pyridines from 2-(trimethylsilyl)-7-chlorofuro[3,2-B]pyridines: to a solution of compound 2-(trimethylsilyl)-7-chlorofuro[3,2-B]pyridine (73 g, 0.323 mol, 1.0 eq.) in tetrahydrofuran (THF, 400 mL) was added an aqueous sodium hydroxide solution ( 300 mL, 4 mol/L). The reaction mixture was stirred at 50 °C for 1 h and then cooled to room temperature and 1000 mL of water was added. The mixture was extracted twice with methyl tert-butyl ether (MTBE). The combined organic layers were washed with saturated saline, dried over anhydrous sodium sulfate, concentrated, and recrystallized from a solvent mixture of ethyl acetate and petroleum ether to give 7-chlorofuro[3,2-b]pyridine as an off-white powder (30 g, 60.5% yield).GC-MS analysis showed the molecular ion peak as 153 (EI). | [References]
