3-Bromofluoranthene is an intermediate in synthesizing 3-Methylfluoranthene (M305315), an environmental pollutant that is an active tumor initiator. | [Synthesis]
Preparation of Compound 2-1: 50 g (247 mmol) of fluoranthene was dissolved in 1 L of nitrobenzene, and a dilute solution consisting of 12 mL (234.7 mmol) of bromine and 200 mL of nitrobenzene was added slowly and dropwise. After the dropwise addition, the reaction mixture was stirred at room temperature for 20 hours. Upon completion of the reaction, 500 mL of saturated aqueous sodium thiosulfate solution was added to quench the reaction. The reaction mixture was extracted with 3 L of ethyl acetate, followed by washing the organic phase with 1 L of distilled water. The organic layer was dried over anhydrous magnesium sulfate and concentrated under reduced pressure to remove the solvent. The crude product was filtered through silica gel column and purified by recrystallization to give the final compound 2-1 (65 g, 94% yield). | [References]
