To a solution of N-((5-bromopyridin-2-yl)methyl)formamide (1.0 g, 4.6 mmol) in toluene (30 mL) was added phosphorus oxychloride (POCl3, 1.0 mL, 10.9 mmol). The reaction mixture was stirred at 103 °C for 2 h. After completion of the reaction, the mixture was concentrated to dryness under reduced pressure. The residue was dissolved in dichloromethane (DCM), washed sequentially with saturated aqueous sodium bicarbonate (NaHCO3) and brine, and the organic phase was dried with anhydrous sodium sulfate (Na2SO4), filtered and concentrated under reduced pressure. The residue was purified by fast column chromatography to afford the target product 6-bromoimidazo[1,5-A]pyridine (0.5 g, 55% yield) as a yellow solid. m/z 197,199 ([M+H]+) was detected by LC-MS. | [References]
