Step 1: Synthesis of 4-(1H-imidazol-2-yl)benzoic acid (248)
To a stirred ammonium hydroxide solution (9 mL) of 4-formylbenzoic acid (2.00 g, 12.3 mmol) was slowly added glyoxal (2.86 mL, 20.0 mmol). The reaction mixture was stirred continuously at room temperature for 16 hours. Upon completion of the reaction, the pH of the reaction mixture was adjusted to 5 with 1N HCl solution.Subsequently, the solvent was removed by rotary evaporator and the residue obtained was ground for 30 min. The milled product was dissolved in water (20 mL) and the solid was collected by filtration to finally obtain 4-(2-imidazolyl)benzoic acid 248 (2.08 g, 83% yield) as a white solid.LRMS (m/z): calculated value 188.1; measured value 189.1. | [References]
