General procedure for the synthesis of 1-methyl-1H-indazole-6-carboxylic acid from methyl 1-methyl-1H-indazole-6-carboxylate and iodomethane: 1H-indazole-6-carboxylic acid methyl ester was prepared with reference to the methodology reported in J. Med. Chem. 2000,43(1),41-58 (Example 12b, p. 49). Subsequently, methyl 1-methyl-1H-indazole-6-carboxylate was obtained in 43% yield under standard alkylation conditions (using sodium hexamethyldisilamate as base, THF as solvent, and iodomethane as alkylation reagent, and refluxing the reaction mixture). Finally, the above esters were treated under standard saponification conditions (1N NaOH aqueous solution) to obtain the target product 1-methyl-1H-indazole-6-carboxylic acid in 96% yield. | [References]
